GENE ENGENEERING-ASSISTED CONSTRUCTION OF ERYTHROMYCIN- PRODUCING STRAINS OF SACCHAROPOLYSPORA ERYTHRAEA K.Pankievich, O.Maryina, S.Zavorotna, V.Fedorenko Ivan Franco National University of Lviv, Hrushevskyi St. 4, UA-79005 Lviv, Ukraine Saccharopolyspora erythraea 5.5 kb DNA fragment containing the genes eryD and eryCI of erythromycin biosynthesis late stages was cloned into shuttle vector pWHM4. Obtained recombinant plasmid was used to transform the protoplasts of eryD-mutant S.erythraea 5002 and ery+ - strains K1 and L5. The renewal of erythromycin biosynthesis by 5002 strain transformants was observed. The high level of antibiotic activity of these transformants corelates with amplification of cloned fragments in their genome. Key words: antibiotic biosynthesis, erythromycin, protoplast transformation, S.erythraea.
